Okuda, Kiyoshi (Department of Biochemistry and Biophysics, University of Hawaii, Honolulu), Gordon C. Edwards, and Theodore Winnick. Biosynthesis of gramicidin and tyrocidine in the Dubos strain of Bacillus brevis. I. Experiments with growing cultures. J. Bacteriol. 85:329-338. 1963.-A simple chromatographic method was developed for the isolation of gramicidin and tyrocidine from tyrothricin of Bacillus brevis. A Tryptone-yeast extract-glucose medium containing mineral salts gave the best yields of peptides in a stationary culture of the organism. The incorporation of suitable C(14)-labeled amino acids into gramicidin and tyrocidine was studied Several analogues of tyrocidine amino acids (beta-hydroxyglutamic acid, pipecolic acid, beta-2-thienylalanine, p-fluorophenylalanine, and phenyl glycine) selectively reduced tyrocidine synthesis, when added to the nutrient medium. At the same time, the production of gramicidin was augmented. Growth and protein synthesis were not affected. Two analogues in isotopic form, beta-2-thienylalanine and isoleucine, were shown to give rise to high degrees of labeling in tyrocidine.

Since the isolation of tyrothricin from cultures of Bacillus brevis (Dubos, 1939), numerous papers have appeared dealing with the chemical and biological properties of this bactericidal agent and its components, gramicidin and tyrocidine (Hotchkiss and Dubos, 1940; Dubos and Hotchkiss, 1941; Tishler, Stokes, Trenner and Conn, 1941; Christensen, Edwards and Piersma, 1941; Lipmann, Hotchkiss and ...

studied soil science. He moved to the Rockefeller Institute because Oswald Avery was seeking an enzyme which would attack the polysaccharide capsule of virulent pneumococci and Dubos offered to find a soil organism which would do so. He succeeded, and went on to seek other antibacterial activities. The search yielded Bacillus brevis, from which Dubos and Rollin Hotchkiss isolated the first clin...

which is synthesised by Bacillus brevis A.T.C.C. 9999 (Laland & Zimmer, 1973). It is synthesized by the enzyme gramicidin S synthetase, which activates and forms thioester bonds with each amino acid before peptide-bond formation (Gevers et al., 1969). Gramicidin S synthetase consists of two enzymes; the smaller, of mol. wt. 1OOOOO (Yamada & Kurahashi, 1969), activates and forms a thioester bond...

Amino acids having a dextro configuration have been found in natural material from two sources.’ Jacobs and Craig obtained d-proline from ergotinine (4) and later other ergot alkaloids, and IvAnovics and Bruckner found d( -)-glutamic acid as the principal constituent of the P-antigen of Bacillus anthracis and other microorganisms of the related Bacillus mesentericus-subtilis group (5). Inasmuch...
متن کاملThe ability of Aneurinibacillus migulanus (Bacillus brevis) to produce the antibiotic gramicidin S is correlated with phenotype variation.
Phenotype instability of bacterial strains can cause significant problems in biotechnological applications, since industrially useful properties may be lost. Here we report such degenerative dissociation for Aneurinibacillus migulanus (formerly known as Bacillus brevis) an established producer of the antimicrobial peptide gramicidin S (GS). Phenotypic variations within and between various strai...
